Annotation description:Treatment with the selective A2A receptor agonist CGS 21680 facilitates the depolarization (20mM K+)-dependent release of radiolabelled glutamate from isolated synaptosomes (Fig 1). The facilitatory effect was blocked by the selective A2A receptor antagonist SCH 5861, but not significantly by the A1 receptor antagonist DPCPX (Fig. 1). Subcellular fractionation of synaptosomes demonstrates the presence of A2A in presynaptic membranes (as well as predominately in postsynaptic membranes; Fig.3) and ICC using an antibody directed against A2A demonstrates a co-localization of A2A and synaptophysin and vGluT1 and 2 immunoreactivity (Fig.4). These data indicate a presynaptic function of A2A receptors in facilitating neurotransmitter release upon binding of adenosine.
