Annotation description:In this paper, Cazares et al demonstrate a novel function of tomosyn-1 despite its effect on synaptic vesicle priming and ternary complex assembly.
The authors monitor the dynamics of distinct synaptic vesicle pools via Vglut1-PhLuorin fluorescence measurements under Wt and tomosyn-1 KD conditions. They demonstrate by quantitative SV pool size measurement the redistribution of vesicles between total releasable pool, TRP (TRP= RRP (readily releasable pool) plus RP (recycling pool)) plus resting pool, ResP (Fig. 1,2). This activity-dependent mechanism of tomosyn-1 consists of clamping of SVs in the ResP and preventing them from transition in the TRP, a function which is regulated by Cdk5-mediated phosphorylation of tomosyn 1. Furthermore, the authors show that tomosyn operates by phosphorylation-dependent association with Rab3a-GTP and synapsin.
