Annotation description:The authors use RNA knockdown for RIM3gamma in cultured neurons to study its role in synapse stability. RIM3 knockdown led to a pronounced reduction in the number of synapsin and PSD95 puncta per um dendrite. In addition, there was a strong reduction in the number of dendritic spines. This suggests that RIM3gamma plays are role in the formation or elimination of synapses.

It should be noted that the knockdown used had more widespread phenotype that include a strong defect in neuronal arborization (Figure 3). It is currently unclear whether the observed phenotype on synapse density is independent of the arborization effect.

The knockdown used was verified with a custom made antibody and western blot analysis (Figure 3A).
