Annotation description:SDS-digested freeze-fracture replica-labeling (SDS-FRL) was used to visualize Cav2.1 in the stratum oriens of the hippocampal CA3 area at high resolution. Cav2.1 was enriched in presynaptic active zones compared to extrasynaptic bouton surfaces. Active zones were identified by clusters of intramembrane particles (IMP) as well co-labeling for the presynaptic active zone protein Rim1/2.

Two Cav2.1 antibodies, one raised in a rabbit (Rb) and another one in a guinea pig (Gp) against different epitopes, labeled the same presynaptic active zone. These antibodies recognized intracellular epitopes on their target protein and thus were visualized by gold particles on the protoplasmic membrane half (P-face). Gold particles labeling the Cav2.1 subunit could not be found in Cav2.1-/- mice.

Additional literature:The distribution of Cav2.1 subunit of the P/Q-type channel was analyzed using highly sensitive SDS-digested freeze-fracture replica labeling in the rat cerebellar Purkinje cells. Localization is found in presynaptic parallel fiber terminals. The specificity of the immuno-gold labeling was confirmed in Cav2.1 KO synapses @ PMID:23426693
