Annotation description:EphB2 is a transmembrane protein of the plasma membrane. In figure 4B, the authors "...purified the synaptosomes of hippocampal tissues and found that Lnx1 could be pulled down by EphB2 antibodies from PSD but not non-PSD fractions...". Thus, antibodies against EphB2 immunoprecipitate the endogenous protein from synaptic fractions. EphB2 is not itself a component of the post-synaptic density (PSD) but interacts with a protein that is at the PSD. In figure 4E, the authors demonstrate "...colocalization of Lnx1 and EphB2 in CA3 pyramidal neurons, immunostaining was used in hippocampal sections and superresolution imaging with stimulated emission depletion (STED) techniques were performed to visualize the localization of these proteins. The STED imaging resolved discrete puncta containing both Lnx1 and EphB2 around the membrane of CA3 neurons". Thus, antibodies against EphB2 co-localize on the membrane of CA3 neurons with a known component of the post-synapse.
