Annotation description:Authors show that NRXN1(SS4+) and NRXN3(SS4+) regulate NMDARs and AMPARs via the cerebellin-dependent activation of GluDs that transduce distinct presynaptic neurexin signals into different postsynaptic receptor responses. The Neurexin alternative splicing site 4 (SS4) is critical to these signaling pathways.

Figure 4,5: Minimal GluD1 (GRID1) and GluD2 (GRID2) constructs containing only their N-terminal cerebellin-binding and C-terminal cytoplasmic domains, joined by an unrelated transmembrane region (of CD4), referred to as GluD1-CD4TMR and GluD2-CD4TMR fully control the levels of NMDA and AMPA receptors. AMPA/NMDA ratio phenotypes of GluD1 knockout1 mice were fully rescued by expressing wildtype GluD1, wildtype GluD2 and the "CD4TMR" constructs thereof.
