Annotation description:Figure 1d-f: immunofluorescence imaging of CNK2 (gene CNKSR2) in cultured primary rat hippocampal neurons at DIV21 showed localization to dendritic spines. Double-labeling showed CNK2 co-localized with PSD-95 (DLG4) and staining was adjacent to presynaptic marker protein synapsin-1 (SYN1).

Based on the high quality imaging and because loss of CNK2 reduced PSD size (Fig. 2) and expression of a CNK2 variant that does not bind to the membrane affected PSD morphology (Fig. 3), this protein was annotated as PSD localized.

This protein was annotated as "extrinsic to the membrane" based on its attachment to the phospholipid membrane via the PH domain and the C-terminal stretch of the PH domain. Authors confirmed this using a EGFP-tagged CNK2 deletion construct that lacks the C-terminal region including the PH domain (Figure S2).
