Annotation description:Using STORM super resolution microscopy and well-validated antibodies, it is demonstrated that RIM1/2 is localized at the active zone, directly opposed to PSD95. RIM1/2 localization is very similar to other well-validated active zone components, including Munc13 and Bassoon (Figure 1). RIM1/2 localization is also shown to correlate with the location of vesicle fusion (Figure 2).

- The RIM antibody (anti-RIM1/2 (1:500; Synaptic Systems No. 140203) used in Fig. 1 has a non-selective RIM1/RIM2 specificity.
- The authors mention identical results with a RIM1 specific antibody ("Similar distribution and nanocluster properties were found using a different antibody targeted towards a separate epitope in RIM1 (Extended Data Fig. 2d)").
- presynaptic localization of overexpressed RIM1 is shown in Fig.2 ("To assess these predictions, we mapped vesicle fusion sites relative to Eos3-tagged RIM1 using sequential PALM-pHuse imaging on the same live boutons (Fig. 2h, Extended Data Fig. 6d, e)")
- Overall these data justify annotation against RIM1.
